摘要:
目的:探讨胆囊癌组织总甲基化水平变化、Bcl-2和p53基因的甲基化水平变化在胆囊癌发病中的 作用。 方法:改进的酶联免疫吸附技术检测45例胆囊癌组织和40例胆囊炎组织基因组总甲基化水平,应用甲 基化特异性PCR技术检测胆囊癌和胆囊炎组织Bcl-2/ p53基因甲基化水平。 结果:胆囊癌组织基因组总甲基化 水平(4.87依1.1%)低于胆囊炎组织基因组总甲基化水平(6.278依0.91%),P<0.05;胆囊癌组织中p53基因甲基 化发生率高于慢性胆囊炎(64%17.5%),P<0.05;胆囊癌组织中Bcl-2基因甲基化发生率与慢性胆囊炎 (17.7%vs15%,P>0.05)无明显差异。 结论:基因组总甲基化水平降低和p53高甲基化可能是胆囊癌发生的分 子机制之一。
Abstract:
To investigate the potential role of methylationmodification(Global DNA methylation/ Bcl-2/ p53)in Gallbladder carcinogenesis. Methods:We collected consecutively gall bladder specimen of 45 Gallbladder carcinoma and 40 Chronic cholecystitis patients. Global DNA methylationwas determinedby aninnovative enzyme-linkedimmunosorbent assay. The methylation-specificpolymerase chain reaction was used to detect methylation status of Bcl-2 and p53. These findings werecomparedbetween45GBCs and40CCs. Results:InGBC,Global DNAmethylationwas lower thaninCC(4. 87依1. 1%VS6. 278依0. 91%,P<0. 05). Thegeneof p53was significantlyhigher frequencies of methylationinGBCcomparedwithCC(64%vs17. 5%,P>0. 05). But,nodifferencein Bcl - 2 (17. 7%vs15%, P> 0. 05). Conclusions: Global DNA hypomethylation and p53 hypermethylationmaybeanimportant molecular mechanisminGBCpathogenesis
